Additive Manufacturing of Poly(Methyl Methacrylate) Biomedical Implants with Dual-Scale Porosity

The development of bone permanent implants with a porous structure favoring their integration with the surrounding tissues is emerging as an attractive field of application of additive manufacturing (AM). This article reports on the investigation of the suitability of a hybrid AM technique, that is, computer-aided wet-spinning (CAWS), to fabricate novel poly(methyl meth- acrylate) (PMMA) constructs as porous implant prototypes. The optimization of the processing parameters to fabricate PMMA samples with a predefined internal porous structure and different external shapes is described. The study demonstrates that tailoring post-processing conditions represents a powerful tool to optimize samples macroscopic aspect, micromorphology, and mechanical properties. In particular, the possibility of obtaining a dual-scale porosity through the integration of the macroporous structure determined by the material lay-down pattern with a submicrometric porosity resulting from the phase inversion process governing polymer solidification, together with the possibility of purifying the employed commercial material from residual monomer during coagulation in ethanol, are reported as note- worthy advantages of CAWS over other AM techniques. A natural progression of this work is the development of relevant complex anatomical prototypes with tailored porosity by processing digital data obtained from computer tomography imaging of bone defects

Supported polymer/lipid hybrid bilayers formation resembles a lipid-like dynamic by reducing the molecular weight of the polymer.

Amphiphilic block copolymers form self-assembled bilayers even in combination with phospholipids. They represent an attractive alternative to native lipid-based membrane systems for supported bilayer formation with applications in biomedical research, sensoring and drug delivery. Their enhanced stability and excellent mechanical properties are linked to their higher molecular weight which generates thicker bilayers. Hypothesis: It is hypothesized that reducing the molecular weight of the polymer facilitates the formation of a thinner, more homogeneous polymer/lipid hybrid bilayer which would benefit the formation of supported bilayers on silicon oxide. Experiment: We investigated hybrid bilayers composed of mixtures of 1-palmitoyl-2-oleoyl-glycero-3-phosphocholine and increasing amounts of a low molecular weight polybutadiene-b-polyethylene oxide copolymer (1050 g/mol). By assessing the bilayer thickness and the molecular packing behavior we sought to demonstrate how reducing the polymer molecular weight increases the tendency to form supported hybrid bilayers in a lipid-like manner. Findings: The formation of a supported hybrid bilayers occurs at polymer contents70 mol% in a lipid-like fashion and is proportional to the cohesive forces between the bilayer components and inversely related to the bilayer hydrophobic core thickness and the extended brush regime of the PEGylated polymeric headgroup

CIRCADIAN RHYTHMS DISRUPTIONS AND EATING DISORDERS: CLINICAL IMPACT AND POSSIBLE PSYCHOPATHOLOGICAL CORRELATES

Background: A link between abnormalities in circadian rhythms and the development of eating disorders was extensively hypothesized, mainly in consideration of the influence of the circadian clock on eating behavior. The present review is aimed at summarizing the evidence about biological rhythms disruptions in eating disorders, possibly clarifying their impact on the psychopathological profile of such patients. Methods: Electronic database MEDLINE/PubMed/Index Medicus was systematically searched for original articles examining the prevalence of circadian rhythms disruptions in eating disorders (anorexia nervosa, bulimia nervosa, binge eating disorder). Results: Studies included in the review confirmed the hypothesis of a high prevalence of circadian disruptions in eating disorders. The analyzed research mainly focused on sleep-wake cycle, rest-activity abnormalities and hormonal secretion, whilst literature about other circadian rhythms was scanty. Altered biological rhythms presented higher association with specific psychopathological features, but such relationship was assessed in few studies. Conclusions: Circadian rhythms disruptions were confirmed to be relevant aspects in the context of eating disorders. Further research is needed in order to clarify the role of biological rhythms in such illnesses, in the attempt to address adjunctive treatment strategies with the possible focus of circadian abnormalities

Tesi

HPLC-HRMS global metabolomics approach for the diagnosis of "olive quick decline syndrome" markers in olive trees leaves

10openInternationalItalian coauthor/editorOlive quick decline syndrome (OQDS) is a multifactorial disease affecting olive plants. The onset of this economically devastating disease has been associated with a Gram-negative plant pathogen called Xylella fastidiosa (Xf). Liquid chromatography separation coupled to high-resolution mass spectrometry detection is one the most widely applied technologies in metabolomics, as it provides a blend of rapid, sensitive, and selective qualitative and quantitative analyses with the ability to identify metabolites. The purpose of this work is the development of a global metabolomics mass spectrometry assay able to identify OQDS molecular markers that could discriminate between healthy (HP) and infected (OP) olive tree leaves. Results obtained via multivariate analysis through an HPLC-ESI HRMS platform (LTQ-Orbitrap from Thermo Scientific) show a clear separation between HP and OP samples. Among the differentially expressed metabolites, 18 different organic compounds highly expressed in the OP group were annotated; results obtained by this metabolomic approach could be used as a fast and reliable method for the biochemical characterization of OQDS and to develop targeted MS approaches for OQDS detection by foliage analysisopenAsteggiano, A.; Franceschi, P.; Zorzi, M.; Aigotti, R.; Dal Bello, F.; Baldassarre, F.; Lops, F.; Carlucci, A.; Medana, C.; Ciccarella, G.Asteggiano, A.; Franceschi, P.; Zorzi, M.; Aigotti, R.; Dal Bello, F.; Baldassarre, F.; Lops, F.; Carlucci, A.; Medana, C.; Ciccarella, G
